Ohh man.. this is such a huge question! And I think it's one of the most important questions asked here in quite a while
Like in all art, there is no right and wrong, only knowledge picked up along the way. And no matter what, that knowledge can always be proven wrong. But I'll try to explain what I've picked up.
A buildup is a urban slang for tension dynamics - this is, the variation of tension over time. Typically we try to build up some tension on the buildup, to later release when the tune comes back in. This means we must have a section that creates tension and another that releases it.

But how can "tense" be defined? If I'm outside and a bomb goes off right next to me, I'll be pretty tense, but Rambo would just call it his morning coffee. So, it depends of the context, on whats been playing, what is playing and what's going to play.

For example, there a friend of mine who produces under DMT Inferno tunes over 320 pbm. There is only a (so distorted it's actually a square wave) kick and a snare, plus sound effects with huge reverb and delay of screams and mayhem in the back. There's no bass, because there's no room for bass.
For this guy to create tension after listening to 3 minutes of this, it's very tricky.. actually he get's it off the other way around, a break releases the tension, and when the song comes back, the world falls on top of you.

Now, trance related, if you put in a KKKKKKKK section it will be more aggressive than what was playing (only a kick every beat), so it could be related to the amount of times something is played. This works charms with the snare (no one ever gets old of a good snare roll). But, you can make this tension you created move, making it come in during time - so, you device to automate the velocity, making the roll start off with 0 volume and end with 80% volume! Yeah, this works, tension is being build! But then the song is cluttered, so you take out the original kick and bass. It's good, but now when the song comes back, it comes back to the same thing. Next you make, say, a new lead to come in when the song comes back. "Hey, I can use this coming in during the build up, introducing the sound before it comes in!", and you start doing it. But, couldn't you use this lead buildup to create even more tension?

Heck yeah you can! If you loop just the first half beat of it during the build, for example, then you'll be playing somethink thats very repetitive, that at the end unfolds itself into the full lead that sounds good. This can build up tension, and you can also loop the leads that were playing - inceptioning yourself to doing this on the whole master track!

Also, The notes you use can build up tension: all notes closer to the root notes are less melodic (forgot the word for it), and notes closer to the center are more melodic - so changing the scale can really do wonders

So you can use sounds, notes, timbres all this in contrast with whats playing to bring in tension. And you can do the same to release it.

That said, filters are the main effects used in buildups, because you can mangle with the timbre of the sounds, chaniging the intensity of their frequencies in a linear way. Taking something out (killing the bass with a Low Pass filter) or bringing some lead in (with a High Pass filter) really does have a huge effect - but consider it the cherry on the cake, not the whole cake. For the remaining effects, you can figure it out on your own if you learn exactly what each does to the signal.

You can use whatever is in your imagination! And that's what makes a buildup huge!

In the psytrance tunes that have been released out to the public Take your favorite tunes, load them um in a DAW, and dissect them. Start cutting it up, try putting kicks and bass and perc and leads ligned up with it, try to recreate the whole tune It's a great experience exercise.

You need to gather more knowledge my friend. My advice over the years: just try to get something done. Give yourself a date, and say: at this date the tune is finished no matter the state. And then make another one!

Peace out!
