Just do whatever necessary to get it to sound the way you want. Why would you ask like we all have a rule book?
  You believe in the users?
Yeah, sure. If I don't have a user, then who wrote me? |

Here's the basics of how I do it.
Get two synth lines playing the same midi file. They don't have to be the same synth patch (I never use the same synth patch).
One is doing the sub, one is doing the mid range.
Start with the mid range line. That mid range synth patch will have a LPF (just like the sub line) doing the filter duties, but it will have a low cut filter on the EQ (200Hz -300Hz I usually go for), along with any other EQing you may need to do to get it sounding the way you want it to.
You dial in your desired filter modulation settings from an ADSR, and since that line is not playing any low end stuff, you are free to up the resonance settings (remember, resonance settings thin out the bottom end, unless the filter cut off is set at the bottom end frequencies). That line is also nice to have a nice wide stereo image, apply a hint of some tight plate reverb and/or tight (stereo) delay with low feedback settings.
Once you get that sorted, go to the sub synth patch. Keep this one mono to have a consistent bottom end from both channels and dont worry about stero image for such low frequencies, the human ear starts loosing directionality below 300Hz.
I keep that patch dry, no tight reverbs (applying reverb to low end parts only muddies up your mix anyway), no delays, no resonance, just a solid bottom end.
Try a hint of ADSR modulation on the filter (24dB/8ve), and instead of using EQ to cut the harmonics, just close the filter enough to get only the low stuff.
If my bassline pattern coincides with the kick, I sidechain the fuck out of the sub bass part (also sidechain the topline, but not as much), but generaly pay attention to kick lenght, bassline note lenght and try low passing your master channel down to 100Hz and have a carefull listen on your bottom end. Its good to hear a full bottom end, yet be able to hear the individual kick and bassline parts, clearly stop at the end of each note. It will help your mix have a defined bottom end over all.
This is how I do it. Talking about it and actually getting it spot on are two different things obviously. My way of thinking makes sense to me, yet I am always striving to put it down to good practise, my self.
Another thing, I never split 16th saw basslines to two parts. Even if I hi pass the top line accordingly, it just sounds iconsistent...to my ears anyway.
Peace out.

in my track 'funk in it' out of 'v/a unrestricted evolution' i have layered 2 basses.
one is only subs (low pass around 100hz) and playing on the base key note to keep the subs steady as possible , while bass guitar sound takes over the mid-bass area and playing on full scale melodic line..
hard sidechain on the sub one.. there are notes on the kick and this why i started the expiriment
the hole thing turned out pritty great , exept maybe some small artifakts that came from not playing the same note on both instruments.
usualy i layer osc's in a synth like z3ta or something , i guess that counts too... mixing saw with square or sine in lower octave turns to diffrent sound then pure one shape. but thats more of a synthesis thing i supose.
