Hello guys it's been a while since i heard this track, and couple days with attempt to make something like this, but without success. xD
move track forward on 4:25 and u will see What i really think. It's sounds like Sylenth1 but i'm not sure.Tell me if i wrong. Cheers

Sounds like a couple of saw waves (maybe a square one in the middle?), not too dissonant, passed through a LP filter with a drive knob, and during that build up the filter's drive is raised, making it scream like that.
If you use some distortion or saturation, you can easily obtain this type of leads. Don't forget to compress, delay and reverb.
Also, when the beat comes back in, you can hear some portamento between notes, that can be achieved with the glide function, or using a pitch bend.
You can make this with a whole whack of different VST's / synths, not just Sylenth1 - the trick is the distortion drive.
