i believe that the best/easier/faster is to work on midi, though when using effects and dynamic processors (eq , compression) is better to work them in audio (maybe i like it), so as developing a track, make your midi patterns and work on the sound inside the vst instrument as best as you can, choose your filters, remove unwanted frequencies, or boost the areas you want, try to fit this specific sound as better you can into your mix and then export/record it to audio, usually 24bit at 44.1Khz
Then having a waveform is more flexible to work on it,, but try to make the best sound to sit on the mix from the virtual synthesizer, the balance is important, how the frequencies are spreading and dont be afraid to leave low or low mid frequencies on some sounds, cause you maybe want them,, in this case you might for example cut them with a filter from the vsti , but maybe that specific filter is poor or you dont like it, my suggestion is leave those frequencies even if they are masking other sounds in your mix like the bass f.e. , and after you make that audio choose a good sounding/ or the proper sounding EQ/Filter for that job and remove or lower them with that one.
it is more like you have to know what your equipment are and WHEN to use them, thats why its a personal aspect of how youll make the sound you want for a mix .. so make the best sound (fat/dry/warm/FATTER ) and try it to stick in your mix having in mind what you can use next to the sound and if that processing is needed and dont overdo it with equalizers and compressors.
one more is never touch the faders of your midi channels inside the sequencer, only the volume inside the instrument to match it to the mix and again before you export it to audio raise the volume, but be careful not to clip, and then lower the audio to the desirable levels back again. hope that helps a bit, but again needs a lot of practice to find out when and why and make your own judgments.. cheers

maybe someone else want to correct my words , if im mistaken at some point but i tried to make a general idea of how to use the instruments .. there are thousands of information and personal opinions on these matters and i tried to be as objective as possible

Do you mean Vst channels?
anyway i often set the channels to "meter input" this way you can see the vst instruments volume and set it as hot as you want.
i often hide my midi channels, but do any one know why the midichannels volume is zero and the panner is set to far left, sometimes when i reset this the volume raise, sometimes nothing happens... btw i use cubase Sx 3

Check the actual values of the MIDI faders and pan controls, don't just look at their positions
And 'never touch the MIDI faders' is bloody good advice.

most important if you want a good sound is to work in a decent studio ,not in a untreated room with lowend monitors,then you can have a good sound with all the vsti |
