I can't figure out how to tune anything very well with it. I have this part that I just want to be tuned to three notes: e, g, and b, but it doesn't really work out well the way i'm doing it. What should my settings look like?

First make sure your sample is tunable, this is something you have to judge by yourself.

Then put in the notes that you want to allow, put autotune speed at 0 or depending on how fast you want the plugin to correct the output tune. Then on the right side you have Pitch Amount, Amplitude Amount and Formant amount. If you want the voice effect put formant on 100 and tweak away!
