1st advice: use Melodyne instead

When to use? When your singer sings out of notes.

I don't have much experience on neither Autotune, nor Melodyne, but have heard some people's reviews that Melodyne does the job more natural.

I've used Melodyne once (as a standalone) and it is really intuitive, natural-sounding (if you are not doing dramatic changes with pitch or whatever) and helpful for fixing the vocal's pitch.

sometimes I like to use something like autotune (logics vocal transformer) on spoken vocal samples in my tracks to make them fit more harmonically.           https://soundcloud.com/hazak

First I'd stick to melodyne.
Second, what about the smurf voices?
Third, that believe song from Cher is so artificial that it is original again, just like Cher. I really like that!

i got em both, i like melodyne better
its great for people who cant sing well,
and u can also do some sphongle vocal efx with it...

autotune is great for the hip hop guys and rnb as well although i heard it on a infected mushroom song cant recall right now, the rappers use it for the t-pain effect.. and lil waynes hit lollipop
cher used aswell like upavas mentioned .

i like melodyne better my choice for creativity,
but autotune i use for all those people who come to my studio for the t-pain effect, easier to work with but limited compared to melodyne hope that helps!
peace
